For the structural proteomics projectofthe hyperthermophilic eubacterium,Aquifex aeolicushas beenselected because of the expectation of stable membrane protein complexes and because of the small size of its genome (less than 1/3 of that ofEscherichia coli) which substantially limits the number of possible membrane protein complexes in this bacterium.The aim of the project is to isolate,tocharacterize and to crystallize as many membrane proteins and membrane protein complexes as possible. Using conventional chromatographic methods we have purified and characterized the NADH quinone oxidoreductase (respiratory complex I), a possible supercomplex consisting of the cytochromebc1complex and of the cytochromecoxidase, the F1Fo-ATP synthase, the cytochromebdoxidaseand sulfid-quinone oxidoreductase, complexes of “hypothetical proteins”. For identification mass spectroscopy is used together with optical spectroscopy and enzymatic assays. A considerable percentage of the isolated protein complexes has yielded crystals.Several crystalsdiffracting X-rays to more than2 Å resolution have been obtained. Two of them had been solved the structures.
